Psilocybin briefly loosens rigid brain networks; therapy may help turn that flexibility into lasting relief.
Psilocybin appears to work through a sequence rather than a single switch: it temporarily loosens rigid network coordination, then altered integration and flexibility may help patients revise entrenched patterns during therapy. Human imaging supports connectivity changes after treatment, while mouse work supplies a possible cellular basis through new and pruned synapses. The bridge from these findings to durable relief remains incomplete.
The lasting effect may begin when old brain routes loosen and new ones become possible.
A mushroom compound changes the brain twice: first by breaking apart familiar network timing, then by helping grow and mature new connection points. That deeper pattern makes depression look less like a fixed chemical shortage than a failure of learning and adaptation. The mouse evidence makes plasticity biologically concrete, but the human path from new spines to durable recovery is still the frontier.
