The Story
Coating mitochondria is a rational delivery-engineering strategy, not yet a demonstrated human therapy. The available evidence supports better uptake or targeting in experimental systems, while clinical relevance remains constrained by mitochondrial quality, tissue delivery, persistence, safety, and the gap between laboratory rescue and durable human benefit.
- A cationic DOTAP/DOPE artificial membrane coated approximately 86% of isolated mitochondria and shifted their surface charge toward positive values, supporting successful coating.
- Surface functionalization can be designed to improve targeting, uptake, and retention in dysfunctional human endothelial-cell models.
- Reviews describe surface modifications, liposomes, and extracellular vesicles as promising ways to improve mitochondrial protection and delivery, but emphasize unresolved translational limitations.

Deeper Insight
The stronger interpretation is that packaging and surface engineering could overcome the central bottleneck: naked mitochondria are fragile and poorly internalized. Encapsulation or coating may preserve function while increasing delivery, making meaningful rescue plausible—but the evidence still depends on whether transferred organelles remain functional and integrate long enough to repair human tissue.
- Membrane coating may increase mitochondrial transfer by improving interactions between isolated mitochondria and recipient cells, addressing a core limitation of naked transplantation.
- Encapsulation and other packaging approaches are being investigated for mitochondrial and Parkinson’s disease models, indicating potential beyond simple uptake enhancement.
- Polymer functionalization has been developed to support cellular transplantation and alter recipient-cell metabolic phenotypes, suggesting coatings can do more than passively protect mitochondria.
